About the Engineer-it Kit™: Refill

This Kit is a refill for our genetic engineering kit for bacteria, the Engineer-it Kit. It does not come with DNA or bacteria (cells). Use DNA and bacteria from a previous Engineer-it Kit, Smell-it Kit, Induce-it Kit, Blue-it Kit, or Heat-it Kit or use DNA you have created yourself. 

This science project refill kit can be used for: 

  • completing more genetic engineering experiments with your existing bacteria and DNA program from the regular Engineer-it Kit, or any of the advanced kits,
  • culturing cells that you have already engineered (e.g. you want to pick a colony and streak some engineered bacteria to grow more)

This refill kit is a perfect way to extend biotechnology experiments at home, in the classroom, the makerspace, community center, or in a local biohacking DIYbio lab!

A course on Udemy guides you through the Engineer-it Kit experience and provides confidence, context, and theory.

What will you learn?

During this biotechnology/genetic engineering experiment, you will learn how to use DNA programs (called plasmids) to make with biology.

The steps for this experiment are:
1. Make selective & non-selective agar,
2. pour Petri dishes,
3. streak bacteria using your own bacteria,
4. make bacteria chemically competent,
5. engineer bacteria using pigment-producing DNA plasmid (using your own DNA),
6. create positive and negative controls,
7. incubate,
8. view/analyze your results!

What's included?

(1) Engineer-it Kit in REFILL Individual size: Sterile water, 6cm Petri dish, LB agar, selection marker, transformation buffer, recovery media, inactivation bag. Since this is a Refill kit, no DNA, bacteria or streaking stencil is included. Use the ones from a previous experiment!

• Individual Kits: An individual kit with the ingredients needed for completing one entire science experience. Great for an individual, family, or a small group — best results using a DNA Playground home or large.

• To pick which antibiotics you need from the drop-down menu:

• If you are using a DNA program and bacteria from the color-pigment Engineer-it kit, you will need the antibiotics Chlor, and no chemicals.
• If you are using a DNA program and bacteria from the Heat-it kit, you will need Chlor and no chemicals
• If you are using a DNA program and bacteria from the Smell-it kit, you will need Chlor and IAA chemical
• If you are using a DNA program and bacteria from the Blue-it kit, you will need Chlor and X-gal chemical
• If you are using a DNA program and bacteria from the Induce-it kit, you will need Amp and IPTG
• If you are using your own DNA program and bacteria, select accordingly


• Instructions: a manual with easy-to-understand vocabulary and step-by-step procedures and an online experiment simulation to practice with, teach you fundamentals, and guide you along the way. See the Resources section below for links

What else is needed?

This kit is a refill and does not include any bacteria or DNA. Use the bacteria and DNA from an Engineer-it Kit or your own lab.

Cleaning and safety supplies are not included in the kit. Necessary safety and cleaning supplies are latex or nitrile gloves, and a chlorinated bleach cleaner or a 10% solution of chlorinated bleach in a spray bottle (you can make this by adding 9 parts water to 1 part bleach in a spray bottle). You can add a Mini Safety Set to your kit which includes gloves and a reusable plastic apron to protect your clothes.

For the best results, and the longest shelf life possible, refrigerate the kit(s) upon arrival. You'll find a "Best by" date on the kit's label. This date assumes you've refrigerated your kit upon arrival.
