Yeast Engineer-it Kit™ for genetic engineering - DNA Arctic Green (beta)

+

Full STEAM ahead with the Yeast Engineer-it Kit. Add coloful Arctic Green DNA to yeast and see the color change!

This kit contains all ingredients needed to grow and engineer yeast with DNA to create a green fluorescent pigment.

Learn genetic engineering hands-on using a Yeast Engineer-it Kit. At the end of this life science project, you'll have grown and engineered yeast to produce something for you -- a colorful bio-pigment! A perfect way for beginners and experts alike to do fun and safe biotechnology experiments at home, in the classroom, the makerspace, community center, or in a local biohacking DIYbio lab!

Just like the original bacteria Engineer-it Kit, the Yeast Engineer-it Kit lets you grow blank yeast and engineer them using DNA plasmids that tell the cells to create a green fluorescent protein and antibiotic resistance enzyme. As you go through the engineering experiment, you’ll see that while there are similarities to engineering E. coli, the transformation protocol is specific to yeast. As you go through the experiment, try to note all the differences. One key idea to note is that the selection uses antibiotic and antibiotic resistance similar to the E. coli kits you’ve done in the past. However, as you’ll see in other Yeast Engineer-it Kits, there are other selection methods!

If you haven't already, you may want to consider starting with the Yeast Canvas Kit, a fun biology-meets-art kit to learn how to grow yeast and make agar Petri dishes. With these basic skills, you can move on to the Yeast Engineer-it Kit with confidence and complete your first genetic engineering experiment!

Unsure what biotechnology and genetic engineering are? Have a look at our short "What is Biotechnology? Genetic Engineering?" post. And remember, no prior knowledge or skills are necessary to get started with Amino Labs beginner's biotechnology kits!

  1. Make selective & non-selective agar,
  2. pour Petri dishes,
  3. streak yeast,
  4. make yeast chemically competent,
  5. engineer yeast using pigment-producing DNA plasmid
  6. create positive and negative controls,
  7. incubate,
  8. View your results

As yeast grows slower than bacteria (36-48+ hours of incubation ), this kit can take up to 1+ week to complete.

During this biotechnology genetic engineering experiment, you will learn how to use DNA programs to make with biology.

  • (1) Engineer-it Kit in Individual or Group size: Sterile water, 6cm Petri dish, agar, selection marker, ‘blank’ yeast stab, transformation buffer, recovery media, positive control yeast stab, DNA plasmid, streaking stencils, inactivation bag.
    • Individual Kits: An individual kit with all the ingredients needed for completing one entire science experience. Great for an individual, family, or a small group — best results using a DNA Playground home or large.
    • Classroom/Group Kits: Eight individually packaged kits with the needed ingredients to complete eight entire science experiences. One shared resource bag containing the yeast, 1 DNA program (plasmid), and inactivation bags, which are shared between all participants. Great for the classroom, advanced AP biology, science fair projects, and iGEM teams. Each of the eight experiments can be done individually or in groups of up to 4.— best results using 2 DNA Playgrounds Large.
  • Blacklight Flashlight (optional add-on): This Arctic Green DNA is only visible under blacklight/UV light. You can add one of our Blacklight Flashlights to your cart when adding your kit, or use one you already have. Classic neon blacklights and UV light boxes to see electrophoresis gels also work.  The Blacklight Flashlight is a 395nm light and requires one AA battery (not included due to shipping regulations).
  • Instructions: a manual with easy-to-understand vocabulary and step-by-step procedures and an online experiment simulation to practice with, teach you fundamentals, and guide you along the way. Available online. See the Resources tab for links!

The kit must be refrigerated upon arrival for longer shelf life.

Necessary equipment: Either the DNA Playground (home or large) or a 30 °C incubator + ice bucket + crushed ice + thermometer + hot water bath.

Blacklight Flashlight, UV light, Blacklight neon or UV lightbox is necessary to see the DNA fluoresce.

Cleaning and safety supplies are not included. These supplies are latex or nitrile gloves, a chlorinated cleaner, or a solution of chlorinated bleach in a spray bottle. For more information about safety, set up and clean up, have a look at the instruction manual(s) for your kit or the safe science page

Our biotechnology education kits for genetic engineering, the Engineer-it Kits, are a general STEM experience that can be used as a hands-on activity to complement various modules from middle school to high school. They allow for scaffolded and project-based learning and are also used in technical colleges, universities, community outreach programs, summer camps, makerspaces, DIY bio & genetic biohacking labs.
Self-contained science project kit: No need to source and buy reagents, chemicals, and consumables. The Engineer-it Kit is a convenient collection of all that is needed to engineer and grow cells in Petri dishes.
All ingredient tubes are color-coded for ease of use. The instruction manuals are a step-by-step illustrated procedure made for beginners with easy to understand language and a glossary for specific technical terms that are part of life science education.
Two convenient kit formats:
  • Individual kit -  One experiment that can be done by individuals, groups, family.
  • Group kit - Eight experiments with one shared DNA program that can be done individually or in groups.
All our biotechnology education kits, including the Engineer-it Kits, are Biosafety Level 1 (BSL1); they do not require any special containment in North America. This means you can do them at home, school, etc. If you are outside of North America, some regulations may apply, please check with your local public health officials. Learn more about safety.
Complements the Life Science in Next Generation Science Standards (NGSS):
