Students will practice streaking techniques to isolate and grow colonies, explore the growth of yeast and E. coli in culture, and apply the scientific method in real-life scenarios. This kit also includes a free presentation on cell culture.

Want to take it a step further? With the optional co-culture add-on, students can mix two cell types and then use their new skills to isolate and obtain pure cultures of each—just like professional microbiologists!

What will you learn?

With this kit, you will:

1. Make selective agar for two different organisms.
2. Pour Petri dishes.
3. Streak bacteria (using various methods to learn streaking methods)
4. Streak yeast (using multiple methods to learn streaking methods)
5. Incubate cells
6. Observe colony formation types, phenotypes, etc.
7. Create hypotheses and practice the scientific method
8. Practice creating streaking patterns to obtain colonies (understanding how to create a lawn and dilute cell concentrations for colonies).

With the co-culture extension kit, you will also:
1. Make selective agar for two different organisms.
2. Pour Petri dishes.
3. Streak bacteria (using various methods to learn streaking methods)
4. Streak yeast (using multiple methods to learn streaking methods)
5. Incubate cells
6. Observe colony formation types, phenotypes, etc.
7. Select colonies with the proper phenotype to isolate the different cell types. Repeating until pure culture is obtained.

What's included?

(1) Streak-it Kit in Individual or Group size: Sterile water, agar, selective antibiotics, Petri dishes (6 cm), loops, bacteria and yeast in stabs, stencils.

• Individual Kits: Contains all the materials needed to perform the entire science experiment once. Perfect for a single participant or a small group looking to share the experience.

• Classroom/Group Kits: Includes four individually packaged kits, each with everything required to complete the full experiment. Ideal for classrooms, science clubs, or larger groups, ensuring every participant has their own set of materials for a hands-on experience.

• Instructions: a manual with easy-to-understand vocabulary and step-by-step procedures and an online experiment simulation to practice with, teach you fundamentals, and guide you along the way. Available online.

What else is needed?

The recommended equipment for this experiment is a 30-37 °C incubator.

This kit is compatible with the DNA Playground's built-in incubator and our Classroom Incubator. You can also build your own DIY incubator by following our tutorial. Have a look in the resource section below for a link.

Cleaning and safety supplies are not included in the kit. Necessary safety and cleaning supplies are latex or nitrile gloves, and a chlorinated bleach cleaner or a 10% solution of chlorinated bleach in a spray bottle (you can make this by adding 9 parts water to 1 part bleach in a spray bottle). You can add a Mini Safety Set to your kit which includes gloves and a reusable plastic apron to protect your clothes.
